MaP555 is a small molecule fluorogenic probe with excellent cell permeability. It is converted from 6 carboxytetramethylrhodamine (6 TAMRA) and has similar spectroscopic properties of 6 TAMRA. MaP555 has an acyl sulfamide structure which favors the spirolactam form, thus has excellent cell permeability. When the probe binds to its cellular targets, MaP555 mainly exists in the fluorescent zwitterion form. The
